Transdermal delivery of cyclosporine A by a newly designed microneedle array device in rats: investigation of the pharmacokinetics and pharmacodynamics. The objective of the present study was to evaluate the pharmacokinetic and pharmacodynamic characteristics of the newly designed microneedle array (MNA) for transdermal delivery of cyclosporine A (CsA) in rats. The formulations of CsA-loaded MNAs, CsA cream, and CsA solution were prepared. The in vitro drug release profiles of the formulations were investigated. The in vivo pharmacokinetics, intradermal/transdermal penetration of CsA-loaded MNAs, and the pharmacodynamic effect of the CsA-loaded MNAs in an experimental allergic conjunctivitis rat model were also evaluated. The in vitro drug release profiles of CsA-loaded MNAs were characterized by a sustained release profile. The pharmacokinetic parameters and drug deposition in skin layers of CsA-loaded MNAs were significantly higher than those of the CsA cream and CsA solution. The CsA-loaded MNAs exhibited lower irritant effects than the CsA cream and CsA solution in an experimental allergic conjunctivitis rat model. The newly designed CsA-loaded MNAs could successfully be applied to the rat skin for the transdermal delivery of CsA without causing irritation.A police officer and three others were shot at the West Oakland home of David Hughes on Tuesday morning. The officer, who was not identified, was hospitalized in serious condition, said Police Chief Howard Jordan. The other three injured officers were in fair condition, he said. The home is near the intersection of 14th and Oak streets, and has been the subject of a police activity since at least last Tuesday, Jordan said. Police received several 911 calls and, after a lengthy police standoff, a SWAT team entered the home about 10 a.m. and found one officer seriously injured, Jordan said. A statement from police said the initial victim was the caller, and that the victims were uninjured. Jordan declined to identify the officer, but said he has been with the force for about a year.
